Patients with Multiple Sclerosis Present an Unaltered Regulatory B Cell Frequency, Phenotype and Function

BACKGROUND: Multiple Sclerosis (MS) is a disease of the Central Nervous System (CNS) classically characterized by the recruitment of T cells into the CNS. However, some data support a role for B cells in its physiopathology. Recently it has emerged that subsets of B cells secreting IL-10 regulate disease symptoms in mouse models of inflammation.
DESIGN/METHODS: Sixty-three MS patients, untreated for at least 3 months and 58 age-matched HV were included in this study. Phenotype and IL-10 secretion by B cells were studied after stimulation with CD40L and ODN. Their suppressive properties on CD4+CD25-T cell proliferation were measured after 3 days of culture.
RESULTS: We report on an absence of differences in the frequencies of the various B cell subsets, as well as the ability of B cells to secrete IL-10 after 5h and 48h of stimulation in MS compared to HV. Analysis of the IL-10+ B cells phenotype revealed mainly a memory CD24+CD27+ phenotype, even though both naive and immature subsets were also able to secrete IL-10. Prestimulated B cells from MS and HV inhibited CD4+CD25- T cell proliferation with a similar efficiency by a contact-dependent mechanism, independently of IL-10 and TGF-?.
CONCLUSIONS: Altogether, our data show that regulatory B cells have conserved frequency and function in the blood of patients with MS, suggesting that they do not contribute to the physiopathology of the disease.
