Title Mediterranean Diet Modifies Risk of Amyotrophic Lateral Sclerosis

Objective

Background BACKGROUND: Dietary habits may influence pathophysiological mechanisms in sporadic ALS, and, since they are modifiable, they may harbor potential preventive interventions.

Design/Methods DESIGN/METHODS: In a population-based case-control study in the Netherlands between 2006 and 2011 we studied the relation between dietary patterns, identified by exploratory factor analysis, and the risk of sporadic ALS, adjusted for confounding factors and corrected for multiple comparison. A food frequency questionnaire was used for data collection.

Results RESULTS: 747 patients and 2,385 controls were included. Two dietary patterns were independently associated with sporadic ALS. A dietary pattern characterized by high intake of total, saturated, and monounsaturated fat, trans fatty acids, and cholesterol and a low intake of alcohol was associated with an increased risk of ALS (p < 0.001). A pattern with high intake of fibre, vitamin C, lycopene, flavonoids, and vegetable protein was associated with a decreased risk of ALS (p = 0.001).

Conclusions CONCLUSIONS: The two identified dietary patterns point to a so-called "Mediterranean diet" and thus provide clues to pathophysiological pathways that might be involved in sporadic ALS. The positive association with fat intake may support a role for insulin resistance, while the negative association with dietary antioxidants supports a role for oxidative stress in the development of sporadic ALS.
