XCiDaBLE: An Observational, Prospective Trial Evaluating Xeomin (IncobotulinumtoxinA) for Cervical Dystonia or Blepharospasm in the United States - Interim Results from the First 145 Subjects with Cervical Dystonia

BACKGROUND: There is a lack of prospectively acquired data on clinicians' "real world" experience with botulinum toxins to treat blepharospasm or CD, especially with newer agents.
DESIGN/METHODS: Subjects are followed for 2 treatment cycles and used Interactive Voice/Web Response. Subject-reported scales include: Cervical Dystonia Impact Profile (CDIP-58) and Patient Global Impression-Severity and -Improvement; Work Productivity and Quality of Life (QoL) is assessed using the SF-12v2, an employment questionnaire and work history.
RESULTS: This interim analysis includes 145 CD subjects (82.3% female, 91.0% white, 77.2% pre-treated with botulinum toxins). At the onset of CD symptoms 73.1% of subjects were employed, but by the start of this study (mean symptom duration 12.6 years) only 44.2% were employed. The mean dose at the first injection was 225 U (naive subjects 159 U and pre-treated subjects 245 U). Four weeks post-injection the majority of subjects reported improvement on the Patient Global Impression. The CDIP-58 total score was significantly improved at four weeks post the first injection compared to baseline (p<0.0001). No new or unexpected adverse events occurred.
CONCLUSIONS: This ongoing study is providing important disease state information for CD patients. In addition, the preliminary results confirm the efficacy and safety in a naturalistic setting of incobotulinumtoxinA that have been previously seen in a double-blind, placebo-controlled study (Comella et al. J Neural Sci. 2011;308(1-2):103-109) and a double-blind active comparator study (Benecke et al. Neurology. 2005;64(11):1949-1951).
