Effects of Intraplantar and Intrathecal Botulinum Toxin Type B on Mono- and Poly-Neuropathic Pain in Mouse

BACKGROUND: Intrathecal (IT) botulinum toxin B (BoNTB) yields a potent analgesia in a murine mononeuropathy (MN) model. Recent work has suggested a possible peripheral action of the BoNTs. We extended our initial work examining the effects of IT and intraplantar (IPLT) BoNTB on allodynia in a MN and a poly neuropathy (PN) mice.
DESIGN/METHODS: An MN was prepared by ligating the L5 spinal nerve root. A PN was made by injecting cisplatin (2.3 mg/kg/day, intraperitoneal;IP) every other day for six times. Saline or BoNTB (up to 1 U) were administered once iplt (unilaterally) or once IT, Mechanical allodynia was assessed periodically before and after IPLT/IT delivery. Motor function of the neuropathic mice was assessed by using the hanging test and grip test.
RESULTS: MN and PN mice showed a prominent allodynia. IPLT BoNTB resulted in a long lasting (up to 21 day) reversal of the allodynia in the PN and MN models. IT BoNTB resulted in a long lasting (up to 21 day) reversal of the allodynia in the PN model. The antiallodynic effects of BoNTB were blocked by prior reduction with dithiothreitol. BoNTB at efficacious doses did not alter motor function.
CONCLUSIONS: IPLT and IT BoNTB yields a persistent attenuation of the allodynia in mouse MN and PN pain models. Wheteer the iplt effects represent a peripheral or central action is unknown.
