Title Methylation Analyses of Stress-Modified Genes in Migraine

Objective To analyze the degree of promoter methyation of genes involved in post-traumatic stress disorder (PTSD) BDNF, CRF, ESR1, GRINB2, GRM1, NR3C1, POMC, SH2D5, SLC6A4, and the recently identified in migraine in an epigenetic study, NPTX2 gene, using a case-control study of migraine patients.

Background In spite of a big effort searching for a "migraine gene", GWAS surveys had yielded poor results. A hypothesis of epigenetic modification of genes that could be involved in migraine pathogenesis takes relevance and a study found a differential methylation degree in migraineurs for NPTX2. As PTSD has a general high prevalence among migraineurs of 22%, we investigated genes previously associated with PTSD and migraine.

Design/Methods

After informed consent, healthy controls (HC), episodic migraine (EM), and chronic migraine (CH) adjusted for sex and age underwent clinical interview for migraine status following IHC-IIIbeta criteria.

Methylation percentage of promoters corresponding to the mentioned genes was abtained using RT-PCR method. Chi2 test, Wilcoxon rank test, Perason and Spearman correlation were used depending on the type of variables and normal distribution.

Results A total of 234 subjects (71 women and 7 men by clinical group) were recruited after sex-and age-matching (39±11y). The degree of methylation of the genes BDNF, CRF, ESR1, GRINB2, GRM1, NPTX2, NR3C1, POMC, SH2D5 y SLC6A4 did not differ among clinical groups. We found strong correlation of age and BDNF, CRF, NPTX2, NRC31 y SLC6A4 genes.

Conclusions In spite of PTSD is frequent among migraineurs, we did not observed association of methylation in genes involved in PTSD and migraine, nor the severity of migraine.
