Methylation Analyses of Stress-Modified Genes in Migraine

To analyze the degree of promoter methyation of genes involved in post-traumatic stress disorder (PTSD) BDNF, CRF, ESR1, GRINB2, GRM1, NR3C1, POMC, SH2D5, SLC6A4, and the recently identified in migraine in an epigenetic study, NPTX2 gene, using a case-control study of migraine patients.
In spite of a big effort searching for a "migraine gene", GWAS surveys had yielded poor results. A hypothesis of epigenetic modification of genes that could be involved in migraine pathogenesis takes relevance and a study found a differential methylation degree in migraineurs for NPTX2. As PTSD has a general high prevalence among migraineurs of 22%, we investigated genes previously associated with PTSD and migraine. 

After informed consent, healthy controls (HC), episodic migraine (EM), and chronic migraine (CH) adjusted for sex and age underwent clinical interview for migraine status following IHC-IIIbeta criteria.

Methylation percentage of promoters corresponding to the mentioned genes was abtained using RT-PCR method. Chi2 test, Wilcoxon rank test, Perason and Spearman correlation were used depending on the type of variables and normal distribution.

A total of 234 subjects (71 women and 7 men by clinical group) were recruited after sex-and age-matching (39±11y).  The degree of methylation of the genes BDNF, CRF, ESR1, GRINB2, GRM1, NPTX2, NR3C1, POMC, SH2D5 y SLC6A4 did not differ among clinical groups. We found strong correlation of age and BDNF, CRF, NPTX2, NRC31 y SLC6A4 genes.
In spite of PTSD is frequent among migraineurs, we did not observed association of methylation in genes involved in PTSD and migraine, nor the severity of migraine.
